Position summary:
Kano is building next-generation genome engineering technologies to enable transformative cell-based therapies. Our platform leverages proprietary circular single-stranded DNA (cssDNA) donor technology to unlock precise, efficient, and scalable genomic insertions. By integrating advances in gene editing, immunology, and cell engineering, Kano is advancing discovery programs at the intersection of immuno-oncology and genetic medicine.

Responsibilities:
Work both independently and as part of a team to design, execute and optimize genomic insertion analysis in primary human cells and cell lines using Kano’s proprietary circular ssDNA donor technology
Detect gene editing outcomes using a variety of methods including flow cytometry, digital PCR, NGS and others as appropriate
Design and produce test constructs to support high-throughput screening efforts, including cloning of plasmids, manufacturing and handling of mRNA, and guide RNA design.
Design, execute, and interpret experiments with molecular biology techniques
Design and optimize fit for purpose analytical methods, including for identification of genomic insertions and subcellular localization of nucleic acids.
Conduct literature and intellectual property reviews and aid in the ideation of potential projects
Write protocols, prepare data presentations for company-wide meetings, maintain a highly organized and detailed electronic laboratory notebook (ELN) and stay up to date with scientific literature
Handle multiple projects effectively, prioritizing tasks, collaborate across teams with ease
Qualifications:
4+ years of professional research experience or PhD in Immunology, Biology, Cell Biology or related fields
Demonstrated experience with CRISPR-based genome editing assay development and interpretation
Experience in molecular cloning techniques for plasmid generation, including PCR, Gibson assembly, restriction cloning, Golden gate assembly, and library generation
Expertise with a variety of molecular and cell biology techniques such as DNA/RNA/protein extraction, sequencing, PCR, ddPCR, ELISA, and western blot
Excellent communication and interpersonal skills, navigates easily working with researchers from all levels of training
Fluency in spoken and written English is required
